Over one battery so it measures 12 V or can it measuring both batteries through the series connection ( 24v ) from the native pole on one battery and the + on second battery?

On the Smart Battery Sense it self, the spec's say: 12/24/48V.

You're free to connect it both on one of the 12V batteries, or on the two 12V batteries in series (24V). In the (advised) 24V option you can connect it to battery 1 negatieve, and battery 2 positive.

The temperature sensor part of the Smart Battery Sense is located inside, so it will only meassure the temperature on which it's mounted.
